Base Concept
If you kill all enemies on a screen that has the "Dungeon Boss" enemy flag checked, all enemies instantly die on screens that belong to the same level number and has "Dungeon Boss" enemy flag checked, so the "One Timed Events" feature can be used to activate a screen's secret from an other screen, being a base utility for complicated story type quests.

Walkthrough
The screen which has the secret triggered will be called Event Screen for now, while the screen where the One Timed Event is activated will be the Trigger Screen. So, the Event and Trigger screens must share the same level number (above 0) but not neccessary for them to be on the same DMap.

The Trigger Screen must have some enemies. Let's use the Trigger enemy now, since it's invisible and untouchable. If you do use the Trigger enemy, place the "Kill All Enemies" item somewhere (preferably its sprite deleted, so it's invisible), let's say, on a combo that looks like a button. Check the "Dungeon Boss" enemy flag (enemy flags appear when pressing F12, too).

If you really placed a button looking combo, set "enemies=>secret" screen flag (F9) and put a secret flag on the button like combo, then set a pressed button as its secret combo. (You should check "no secret sounds" screen flag, so the secret sound won't play every time you reenter this screen.) Now, on the Event screen, set the secret that you want to activate (make sure it can't be activated on the screen, itself, so place only secret flags "16-32", no trigger flags), check the "dungeon boss" enemy flag (F12), choose the Trigger as an enemy, don't forget to check the "enemies=>secret" screen flag and preferably check the "no secret sounds" screen flag (F9). If you do all of this, the following happens: the player steps on the button in the Trigger screen, it gets pressed and the secret is activated in the Event screen.

Examples
With the above walkthrough, you can make it so that the player steps on a button in one screen and a gate opens in other screen. You can make it so that you kill a gleeok in one screen and so a gleeok statue disappears from the other screen. You can make it so that the player can choose between the two rooms and killing all enemies in one of them makes the enemies disappear in the other one. You can make it so that the player steps on a button and a hard enemy disappears from the other room. You can make it so that an invisible warp appears on the Event screen and it takes the player to a screen that is similar to the other behind the Event screen, just the passage is unblocked. The only limitation is your imagination.

Warning
The "Kill All Enemies" item doesn't work if it is placed near the side of a dungeon DMap's screen, nor when the player teleports into it. "Guy"s also count as enemies. The Trigger enemy is invisible, but it still requires a place to appear somewhere: have huge open walkable area, or set the enemy flag to a walkable combo. If there are other enemies besides the Trigger enemy, the Trigger enemy must be the first in the list.
